Epson 4870 with medium format 645 transparencies

I'm trying to scan 645 slides with an Epson 4870, using the medium
format film holder. These are cut transparencies; not in a strip.
Will it work? I'm not sure what settings to use but it is not
recognizing the images. Is there some trick to it? I can fit a total
of 6 of them in the film holder that they provide. I just got this
scanner and don't understand what the "Target size" is or whether this
has anything to do with my problem. Thanks!

Go into Professional or Home mode, click on the small arrow next to the
Preview button, choose Normal instead of Thumbnail, draw your marquees
manually for each strip. You can configure it to scan multiple marquees in
one batch.
