So, after unchecking the "disabled 3rd party browser *extensions*", and
after restarting IE (which means you close *ALL* instances of IE, even those
without windows so be sure to check Task Manager's Process tab for any
iexplore.exe processes), this option is getting checked again? Sounds like
you have some nastyware installed.
You could use BHO Demon or Spybot's tools to see what browser helper objects
(and ActiveX components) have been installed. Another way to modify IE is
to add registry keys under:
HKEY_CURRENT_USER\Software\Microsoft\Internet Explorer\Toolbar

will actually affect a change in behavior (other than to add the toolbar).
Some viruses, trojans, and spyware will try to protect itself. It is got
disabled, it couldn't do its nasty. Have you ran a full scan using a
recently updated anti-virus product? Have you ran Ad-Aware, Spybot, and
CWShredder to make sure you aren't infected?

To see the registry key that gets modified. Try exiting all IE instances,
change the data item to disable extensions, and then see if it gets enabled
after starting IE.
