disk cleanup

I have windows xp, lately my computer has been slowing down, so I went to use disk cleanup after running a virus scan, and the disk cleanup will not work. I went ahead and defragged, which it said I didn't need to do but felt it was smart. But I still cannot get the disk cleanup to work and don't know what to do, can someone help me please.
