Enter your values in A1 and A2 as a real date, then a space character, then the
time:

05/03/2007 07:21:00 AM

After you've entered the date and time, you can format the cells the way you
like:

hh:mm:ss mm/dd/yyyy
(for instance)
One way:

A1: 03:30:00 01/25/2007
A2: 05:30:00 01/27/2007
A3: =A2-A1

returns

50:00:00

when formatted as [hh]:mm:ss

As always, if you are doing a calculation it doesn't matter what format the
source cells are using to *display* the data; what does matter is that the
data values in the cells are stored as Excel's date and time. It won't work
if your input data are text strings.
