Can't defrag large file

I have downloaded footage from a digital video camera through Movie Maker 2
thus creating a very large file (11Gb). Now on running defrag it seems
incapable of defragging this file. I need this source file for a video
project I have spent many weeks on (it can't be deleted). Does anyone have a
solution?

How large is the partition in which it is located and how much free
space is there in that partition. Does your partition also contain
your Windows operating system?

Try defragmenting with this and see if it helps or try the trial
version of a third party tool.

If you have another partition, try moving the file to it, and then move it
back to its original location. Also defrag the original partition (to clean
it up) before moving the file back.
