Application won't run.

I have an old DOS based application (DataFlex) that I run on my laptop and workstation. The program does and alwasys has run fine on the laptop however if I try and run it on the workstation I get the following error message

16 bit MS-DOS Subsystem

c:\docume~1\Dennis~1\Locals~1\flex.exe
c:\windows\system32\AUTOEXEC.NT. The system file is not suitable for running MS-DOS and Microsoft Windows applications. Choose 'Close to terminate the application.

Both machines are running Windows XP Professional.

Any idea why one works and the other does not?
