Access to folder 'access is denied"

I had to re-image my machine. Two physical drives; I formatted and
reinstalled the OS on the primary drive. The secondary drive remains intact
with the current file structure. The primary is formatted FAT32 (bad choice
for many reasons) and the secondary NTFS. I can access all folders and
files on the secondary drive except those in my user directory. "access is
denied". Is there a way to change at the "system level" the permissions of
this folder to allow access?
