I have my C: drive is the master and a second drive (D:) is the slave on the
primary cable where I clone my C: drive to. This clone works flawlessly

I plan to add another drive which will be the slave position on the
secondary cable. Will a clone of drive C: on this additional drive result
in a usable bootable copy from this secondary cable location?

Should be fine. It just depends on what the bios can boot from
and any decent modern bios should be able to do that fine.
