One Scanner Two Color Profiles

With a flatbed scanner such as the Epson 4870 (and all other net/flatbed
scanners for that matter) do I need one profile for reflective mode and one
profile for transparency mode? How profoundly do the two different light
sources affect calibration or is the CCD sensor the more relevant factor in
the process? Also, will using an IT8 target slide (e.g. Wolf's Faust's
Ektachrome) for calibrating transparency allow me accurate colors with
negative films too?

Not sure, but I believe the different light sources affect
calibration/profiling more.

No - the natural orange colorcast of negative films will be an overreaching
factor.
