How to logon as administrator

Have a virus problem with our computor. Have isolated the problem file, but
am unable to get access to it. Can't delete it or rename it. I have tried to
use the security tab to authorize the user but it won't accept the change. I
suppose I need to log on at the outset as administrator, but I don't know
how that is done at startup. Have windows professional on the computer.
