Does computer have to be ON for a message rule to work?

I guess the subject line says it all? I set a message rule in Windows Mail
(on my home desktop) to forward emails from a certain person to my work email
so I can receive it while at work.
My question is: If my home computer is OFF, will I get the email at work? Or
do I have to leave my home computer turned ON all day while I'm not there?

No. The mail client has to be open. You might try and look if your ISP has
webmail on which you could set up a rule...
