If you Google for "letter of intent," you'll find that it's a pretty generic
term. If you Google for "letter of intent" job opening, you'll get a more
focused selection of links, but again the term is used fairly generically.
For example,
http://envision.ca/templates/resources.asp?ID=3156 gives
guidelines for a letter of intent with regard to applying for funding for a
nonprofit organization. There's a model LOI for another purpose at the end
of
www.wsgr.com/PDFSearch/risky.pdf . There's a sample "cover letter or
letter of intent" in
http://cf.linnbenton.edu/depts/careerservices/
upload/bluebook.pdf. In some cases the LOI is the letter in which the
employer offers you a job. I think you could just write a letter similar to
what you would write in applying from outside, except that presumably your
boss knows your qualifications, so the submission of a formal résumé,
references, etc., would not be required.

I was told to submit a Letter of intent and am unsure of the proper
formatting for such a letter. I find many templates, samples, and
suggestions on the structure and format for a standard cover letter,
however, nothing for internal positions. Of course, to a degree, one can
say exactly what one would say to the employer directly, however, if the
boss is asking for this kind of letter, I figure there must likely be a
template or two out there.
