cannot lock current drive,Help

I'm running Windows Xp Home Edition and when I try to run Chkdsk /f from the command prompt I
get the error "The type of system is Fat32 Cannot lock
current drive.""Chkdsk canot run because the volume is
in use by another process. Would you like to schedule
this to run at start up?" Type Yes or No? I type in yes and Restart and it stills tells me the same thing
I can't run defrag cause it tells me i have problems,so I have no idea how to fix this.please help me....Thank
