Install it and then it should be listed under Start -> Programs ->
PowerToys...

TweakUI is a .exe file that you download. Once you have downloaded it double
click on the TweakUI.exe file and it will install.
After it has been installed you start the program manually.

JS. I found tweakui.exe in programs, but i am not sure how to use or open a
file manually. please advise

Just click on it. Then click on "My Computer" and then "Special
Folders". And then select My Music from the drop down menu and tell it
where to go too. Easy eh? I'm using an older version (2.10.0.0) and
hopefully it is still the same.

Sorry, I misunderstood, as it looks like you have already installed TweakUI
if it's listed in Start/All Programs/Powertoys for Windows XP/TweakUI. If
this is the case then just launch it as you would any other program.

Why did you download it if you

1) don't know how to open a program, and

2) don't know how to use it when you finally figure out how to open
it (click or double-click it!)?
