How do I wake up my computer without turning it off and on. Pushing the
power button on my desktop or clicking ht mouse does not work.

I have a Gateway GT5657E with Vista.

I think it depends on what type of PC you have, and the details of
your sleep mode setting. I can wake mine by moving the mouse
around.

On my Dell, the Mouse itself goes to sleep and clicking L/R buttons
doesn't wake it except the centre button under the wheel. (This is
a radio mouse so it dozes off quite readily!)
 
Go to Advance Power Settings.
Set hard drive to turn off a minute or 2 before sleep.
Set Hybrid and Hibernate modes to "Off"
Under USB settings select "Disable"
Under Multimedia select "Allow ....."

In sleep mode, moving the mouse usually works. If not, try hitting the
Shift key on your keyboard.
