It always is a good idea to post the version of PowerPoint you are using,
since answers vary from version to version.
Did you move the sound files to a new directory? Most sound files are
linked, not embedded, so moving the source may break the link. See this for

What media programs have you added recently? QuickTime? Winamp?
RealPlayer?
In some versions of PowerPoint, the sounds are played by the Windows MCI
program and the settings for this MCI program can be corrupted by installing
media programs. Check this link to try to track down where your settings
may have gone astray.
