Could be any number of reasons ranging from low free space to filesystem corruption or lack of admin privileges. Without the error message, it's impossible to know.
If it's low free space, then clear up some space, or try another defragger. You can download free evaluation versions of the popular commercial utilities, so head over to the Diskeeper website and get a copy of DK2009. It's a great program; run it in automatic mode, and it should defrag even with less than 5% space.
