Help with Hard Drive positioning

I bought a 120mm fan to use for front intake. My hard drive holder is
positioned right in front of the fan (the HDD is in a horizontal position).

I have a 4 slots in this HDD holder. The top and bottom slots don't block
airlow, but the middle 2 do.

My question: should I put the HDD in one of the middle slots for extra hard
drive cooling? Or would this restrict overall airflow for the other
components?

Yes.
 
Arrange it so you get the best airflow past the HD. You'll get better
airflow into/through the case. Even if the fan is not blowing diectly
onto the drive, you're exchanging the air around it so it'll still cool
the drive.
Not that it matter much but in case you've never noticed, the center of
the fan isn't where the most airflow/velocity is. It's near the edges.

I'd not worry about HD cooling - worry about CPU cooling; therefore, vent
the case as best as possible.
