Can't delete a file

I've somehow corrupted an 800 MB video file in a presentation so that I can
no longer edit of delete the file. I have a backup but need to be able to
reclaim the space. I've tried rebooting and shutting down all other apps
but it still tells me the file is in use.

Can someone tell me how to unlock this file so I can get back 800 MB?
