Sure, if it's formatted NTFS. Go to the root of the drive and open the
properties and select the security tab and remove the Users group from
the list of accounts with permissions. You may also have to remove the
Everyone group, if present.

If XP Pro, disable simple file sharing to see the security tab. In Home
Edition, boot in safe mode to see the security tab.
