16 Bit System 32 message

I am trying to install a new program on my computer but I keep getting the
following message: "C:\windows\system32\autoexec.nt. The system file is not
suitable for running MS-DOS and microsoft window application." How do I
correct this? This program has been successfully installed before on the same
computer

Contradiction..

"trying to install a new program on my computer"
and
"This program has been successfully installed before on the same computer"

Was it on the same Operating System?
What appl,ication is it?
Have you tried installing/running it in Compatability mode(s)?
