Why fill out the Relationship screen?

What is the advantage in defining relationships in the Relationship screen if
I can define the relationships I need right in the queries when I need them?

Because those lines you create in queries are NOT relationships. Relationships
impose rules upon the data that can be saved in tables. The join lines in
queries simply dictate what data a query will output or act upon.
