Windows 2000 Pro Defrag prob

I' use a PC with Windows 2000 pro, my problem is that I can not defrag the
HDD.
I need to run CHKDSK first, if I set CHKDSK from within Windows - when I
restart the PC CHKDSK is unable to run as it can not get exclusive access to
the HDD.

I have restarted the PC in Safe Mode with command prompt, I can run CHKDSK
but not with the /F switch.
CHKDSK still can not get exclusive access to the HDD.
How can I find out what processes are still runing at this point.
