Windows XP home - NTFS files - cannot defrag - chkdsk and defrag

When I try to defrag my c: drive I get the following message:

Disk Defragmenter has detected the chksdk is scheduled to run on
the
volume: (C):

Please run chkdsk /f.


When I run chkdsk /f I get the message:

Cannot lock the current drive

Chkdsk cannot run because the volume is in use by another process.
Would
you like to schedule this volume to be checked the next time the
system
restarts? (Y/N)

No matter if I type yes or no when the systme restarts I still cannot run a
disk defrag.

I am unable to perform a back up as well

Does chkdsk run after you type "Yes"?
Try the drive Manufacturer's website and download/run Their drive test
utility.
