My definitions are 2 weeks old and I have automatic updates turned on before
scans. I did a full scan a week ago. When trying to do the update
manually, I get an error Windows Defender unable to complete the update
0x80070005 access denied. What do you recommend I do?

At the top of this page there's a box that says Search For:
and another that says In:
Type or paste 0x80070005 in the Search For: box, and pick General,
Announcements, and Signatures In the drop down menu on the other box.
