should you set up the tcp/ip configuration for Preferred DNS Server and
Alternate DNS server on them? Should each server only point to itself
for preferred DNS server and not set an Alternate DNS server?

No.

You should generally use both. Under certain conditions the "other"
should even be first but generally go with the "most efficient" which
is the same server first, other DNS second.
