How do I designate a shared folder on a pc as a drive letter. I can easily
designate a drive letter from another workstation as a mapped drive letter.
But, on the pc that is sharing the folder, I want to designate that same
folder with a drive letter, so no matter which workstation a user is using,
they know to save date to the designated driver letter.

See the Subst command in Windows Help.

Unless one is using the Command Prompt, how does that help? Is there a way
to get it into the system variables? I can't see how else it could work in
windows.

You may use a batch file to load it using a shortcut either on the
desktop or in the Programs, Startup folder. That letter is then
used/available in the GUI.
