Code behind vs one file coding

hi, I realized that there are two ways to create a web
page in asp.net . either put allthe code in one page and
use the tags to separate the server code from the client
code. or use what vs.net offers, code behind aspx and
aspx.vb

The question is what is better to develop with and what if
I have an asp page that was created in vs.net with code
behind and I need to make it one file.
