I've started getting the message "Could not find web
server (website URL) on port 80..." when trying to update
the sites theme. The site is rather large (upwards of
500-600 photos and 50-60 pages). I'm, of course, already
logged into the site when the updates are being made.

The server may be having problems, ask them, if the upload
starts but ends at somepoint it may be on your end. Right
click most of your files and select don't publish then try
to upload. If this works, just upload a few at a time.
once the site is publish you will need to add a few files
at a time. Publish as often as you can to avoid large
updates.
