Try what Gordon said. Also note that the command-line version of Disk Defragmeter (C:\Windows\System32\defrag.exe) still exists in Windows Vista. You'll need to open an elevated Command Prompt and run it. Type "defrag.exe /?" to know the supported arguments.
If that does not work, check if you have the file defrag.exe in your Windows\System32 folder.
