Therefore, I have used a rule to assign a meeting appointment to the user's calendar and a receipt to ack the meeting. Is there a way of looking a meeting-rooms mailbox to check whether a recipient is available on their calendar and then scheduling the meeting?

Yes, but it would take custom code, not a simple rule.
