It'd help if you said what board?
Most modern boards will accept memory in any slot, in any order, but some
slightly older generation boards use the memory in the 'first' slot, to
set the memory timings, and therefore require the slots to be populated in
order.

One in perhaps 50000hours, may reflect just a cosmic ray 'hit', rather
than a physical problem (which is why ECC is more reliable), but except
for this, there should be zero memory errors.
