What is the optimal way to set this drive up? Ultimately it will be
strictly used for video editing so I am assuming the more available space
the better...

Any advice from any others who use these large drives for video editing?

Thanks!

Mine is divided into two partitions, one at 20gb for the O/S and the other
at 180gb for just video files. The other two hard drives are 80gb and 120gb
respectively. I've had files as big as 80gb on the largest drive at times
while I'm editing, so I really need those large partitions.
