is there any threat to the 16 bit applications we are running on WINDOWS XP
PRO -SP2
in near feature with any kind of updates/ auto updates micorosoft is pushing
or if we install sp3

You can always use a test machine and see if the updates will cause an issue
if it didn't then you are safe.
Note the machine need to be the same settings and hardware.

There is absolutely no way to be sure unless you test this. If it's
important to you, set up an inexpensive test system and try it. Don't take
anyone's word that it does or doesn't work.
